Effective date : Many hormones, neurotransmitters, and biologically active substances control, regulate, or adjust the functions of living organisms through specific receptors located on cell membranes. Many of these receptors mediate the transmission of intracellular signals by activating guanine nucleotide binding proteins G proteins to which the receptor binds. The binding of a specific signaling molecule to GPCR can cause a change in the conformation of the receptor, resulting in a form capable of binding to and activating the G-protein, thereby initiating a cascade of intracellular events, which ultimately leads to a biological response. Typically, GPCRs interact with G-proteins to regulate the synthesis of intracellular secondary messengers such as cAMP, inositol phosphates, diacylglycerol and calcium ions.
